Enhancers are traditionally considered self-sufficient in upregulating gene bursting. However, our previous observations indicate that the Sox2 gene promoter, despite its proximity to a super-enhancer, requires a nearby diffraction-sized transcriptional condensate for enhanced bursting. Based on these observations, we proposed a mechanism known as the three-way kissing model. By employing advanced super-resolution microscopy, genomics, and transcriptomics, I aim to develop methods and techniques to assess the universality of the three-way kissing model and understand the role of these condensates in enhancing gene bursting.
